Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

TPS1 encodes the synthase subunit of the trehalose-6-phosphate synthase/phosphatase complex and is primarily localized to the cytoplasm, where it can function within the multi-subunit complex and also be detected in a monomeric form. The canonical protein is ~495 amino acids (~56 kDa) and catalyzes formation of trehalose-6-phosphate from glucose-6-phosphate and UDP-glucose (i.e., a UDP-glucose-dependent trehalose-6-phosphate synthase activity), providing the committed step toward trehalose production. Biologically, TPS1 supports accumulation of trehalose, a key storage and stress-protective carbohydrate, and its activity is required for robust stress adaptation phenotypes such as osmotic and desiccation tolerance; loss of TPS1 abolishes measurable trehalose and trehalose-6-phosphate synthase activity.
