TRPM6 is predominantly expressed in the kidney and colon, and encodes a protein containing an ion channel domain and a protein kinase domain. TRPM6 is crucial for magnesium homeostasis, and plays an essential role in epithelial magnesium transport and in the active magnesium absorption in the gut and kidney. Further, TRPM6 is an essential ion channel and serine/threonine-protein kinase, and is crucial for magnesium homeostasis. TRPM6 has an important role in epithelial magnesium transport and in the active magnesium absorption in the gut and kidney. Isoforms of the type M6-kinase lack the ion channel region. Mutations in TRPM6 are associated with hypomagnesemia with secondary hypocalcemia. Alternatively spliced transcript variants encoding different isoforms have been noted for this gene. TRPC7 is thought to form a receptor-activated non-selective calcium permeant cation channel. TRPC7 is probably is operated by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases or G-protein coupled receptors. TRPC7 is also activated by diacylglycerol (DAG). Further, TRPC7 may also be activated by intracellular calcium store depletion.
