TTI2 Antibody (PA5-98544) in ICC/IF

Immunofluorescent analysis of TTI2 in A549 cells using a TTI2 polyclonal antibody (Product # PA5-98544) at a dilution of 1:33. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated Goat Anti-Rabbit IgG(H+L). Cells were counter-stained with DAPI.

The DNA replication (origin) licensing system is necessary for duplicating the genome in each cell cycle. Geminin is a protein that prevents DNA replication by blocking the MCM complex incorporation into prereplication complex (pre-RC). It is absent during G1 phase and accumulates during S, G2, and M phases of mitosis. It is then degraded during the metaphase-anaphase trasition allowing replication to occur during subsequent cycles. Geminin has also been implicated in maintaining integrity of the male germline genome.
