TYLCV C3 is a complementary-sense open reading frame encoded on the circular, single-stranded DNA genome of tomato yellow leaf curl virus (a monopartite begomovirus), positioned among the cluster of complementary-strand genes that overlap and are transcribed in opposition to the virion-sense genes. The C3 locus encodes a small replication enhancer protein (REn/C3) that lacks obvious enzymatic motifs but functions through protein-protein interactions, including self-association and binding to the viral replication initiator protein C1/Rep, thereby boosting viral DNA accumulation during infection. Mechanistically, C3 promotes efficient geminiviral DNA replication in plant cells by engaging host replication machinery: it interacts with the host processivity factor PCNA and has been linked to modulation of cell-cycle control via interaction with retinoblastoma-related protein, consistent with a role in creating a cellular environment favorable for rolling-circle replication and complementary-strand synthesis.
