TYLCV C4 is a complementary-sense open reading frame in the monopartite circular ssDNA genome of Tomato yellow leaf curl virus (family Geminiviridae, genus Begomovirus), positioned among the virus-encoded C genes and transcribed from the complementary strand; it encodes a small multifunctional protein expressed in infected plant cells. C4 acts as a pathogenicity determinant that contributes to disease symptom development and severity and is required for efficient systemic spread in tomato, as disruption of C4 markedly attenuates symptoms and compromises long-distance movement in the natural host while still permitting replication in single cells. Beyond its role in movement and symptomatology, C4 broadly perturbs host signaling and defense pathways by targeting host factors; reported activities include modulation of developmental gene expression programs associated with leaf-curl phenotypes and interaction with multiple plant receptor-like kinases, consistent with C4 functioning as a virulence effector that rewires host regulatory networks to favor infection and systemic colonization.
