Themis Antibody (MA5-50142) in ICC/IF
Immunocytochemistry analysis of HepG2 with Themis monoclonal antibody (Product # MA5-50142) at 1:40, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 503-congugated AffiniPure Goat Anti-Rabbit IgG (H+L).

Themis is a cytoplasmic protein that is essential for positive selection and the development of single-positive thymocytes. Highest levels of expression are found in CD4+CD8+ double-positive thymocytes, with lower expression found in mature T cells. In addition to Grb1, Themis has been found to associate with PLC gamma 1 and LAT and is quickly phosphorylated by SLP-76 following TCR signaling. There are 5 isoforms of mouse Themis that are produced as a result of alternative splicing.
