Toc75 is an essential Omp85-family beta-barrel protein embedded in the chloroplast outer envelope membrane, where it constitutes the core protein-conducting pore of the TOC (translocon at the outer chloroplast membrane) import machinery for nuclear-encoded preproteins. It is synthesized as a precursor that is sorted to the outer envelope, and mature Toc75 is organized as an N-terminal soluble region exposed to the intermembrane space containing three polypeptide transport-associated (POTRA) domains, followed by a C-terminal membrane-integrated beta-barrel predicted to comprise 16 transmembrane beta strands that form the translocation channel. Functionally, Toc75 cooperates with TOC receptor GTPases (e.g., Toc34 and Toc159-family receptors) to receive cytosolic preproteins bearing chloroplast transit peptides and to mediate their passage across the outer membrane en route to downstream TIC components at the inner envelope; biophysical and genetic data support its role as a selective, gated pore that can interact with transit peptides and is required for efficient chloroplast protein import and organelle biogenesis.
