U1A (encoded by SNRPA) is a predominantly nuclear, spliceosome-associated RNA-binding protein enriched in nucleoplasm/nuclear speckles, where it forms part of the U1 small nuclear ribonucleoprotein particle that recognizes 5-prime splice sites during pre-mRNA splicing. The protein is built around two RNA recognition motif (RRM) domains, with an N-terminal RRM that binds with high affinity and sequence specificity to stem-loop II of U1 snRNA, while the C-terminal RRM is less clearly assigned to RNA binding and is thought to support additional protein-protein interactions within RNA-processing assemblies; regulatory elements including a nuclear localization signal contribute to its nuclear accumulation. Functionally, U1A stabilizes and positions U1 snRNP on nascent transcripts to promote intron removal and has a well-defined autoregulation mechanism in which excess U1A binds a conserved element in its own 3-prime untranslated region and inhibits polyadenylation of its pre-mRNA via a dimerization-dependent process, linking splicing-factor availability to 3-prime end formation and transcript output.
