UBC13 (UBE2N) is a canonical E2 that forms an obligate heterodimer with a catalytically inactive E2-variant partner (e.g., MMS2 or UEV1A) to assemble Lys63-linked polyubiquitin chains that function primarily as non-proteolytic signals, with documented roles separating DNA damage tolerance/repair versus NF-kappaB signaling depending on the specific UEV partner. UBC14 (UBE2E1) is an E2 with a UBC catalytic domain and multiple splice isoforms; it participates in ubiquitin transfer reactions implicated in nuclear protein regulation, including histone H2A Lys119 monoubiquitination with PRC1-related activity, and shows regulated nucleo-cytoplasmic distribution linked to control of ubiquitination-dependent stability of key signaling proteins.
