Urocortin 3 (UCN3) is a member of the corticotropin-releasing factor (CRF) family, encoded by the UCN3 gene, and is primarily involved in stress response and energy balance regulation. UCN3 is predominantly expressed in the hypothalamus and certain peripheral tissues, such as pancreatic islets, where it plays significant roles in modulating anxiety-related behaviors and metabolism. This neuropeptide functions by binding to CRF2 receptors, exerting effects on glucose metabolism and insulin secretion. UCN3 has been implicated in protecting the heart from ischemic damage and modulating the cardiovascular system's response to stress, highlighting its potential in treating cardiac disorders. Additionally, it influences nutrient partitioning and energy homeostasis, with expression levels affected by nutritional status. Research indicates that UCN3 deficiency may lead to impaired glucose tolerance and altered energy expenditure, demonstrating its importance in metabolic health. Furthermore, UCN3 has been associated with the regulation of neuroendocrine signaling pathways, providing insights into therapeutic strategies for metabolic and stress-related disorders.
