V-ATPase a3 (encoded by TCIRG1) is a membrane-embedded "a" subunit of the V0 sector of the vacuolar-type H+ ATPase (V-ATPase), a rotary proton pump that acidifies intracellular compartments and, in specialized cells, the extracellular space adjacent to the plasma membrane. In osteoclasts, a3-containing V-ATPase is enriched on secretory lysosomes and is delivered to the ruffled border membrane, where it drives proton secretion to acidify the resorption lacuna required for bone demineralization and matrix degradation; in addition, a3 contributes to secretory lysosome trafficking by recruiting the small GTPase Rab7 to lysosomal membranes. Structurally, a3 is a large integral membrane protein forming part of the V0 proton-translocation machinery, with a substantial cytosolic N-terminal domain implicated in regulatory/trafficking interactions and a multi-pass C-terminal transmembrane domain that participates in the membrane channel interface with the proteolipid ring. Loss-of-function variants disrupt organelle/extracellular acidification and osteoclast polarity/ruffled border formation, consistent with the established association of TCIRG1 defects with infantile malignant osteopetrosis.
