ZCP1 (zona pellucida glycoprotein 1; ZP1) encodes a secreted, extracellular matrix glycoprotein that is synthesized by growing oocytes and incorporated into the zona pellucida surrounding the oocyte and early embryo. The gene produces multiple transcripts and a protein characterized by an N-terminal signal peptide for secretion and a conserved zona pellucida (ZP) domain (approximately 270 amino acids with conserved cysteines forming intramolecular disulfide bonds) that supports polymerization/assembly with other ZP proteins; in the assembled matrix, ZP1 functions primarily as a structural cross-linker that stabilizes and reinforces the fibrillar scaffold built largely from ZP2-ZP3, thereby maintaining zona pellucida integrity required for normal fertilization and preimplantation development. Experimental and clinical genetics evidence links loss-of-function or damaging variants in ZP1 to defective or absent zona pellucida, oocyte maturation defects, and female infertility, consistent with its essential role in constructing a functional oocyte coat.
