cpCK2 encodes a nucleus-encoded, chloroplast-targeted serine/threonine protein kinase that localizes predominantly to the chloroplast stroma, where it functions as a CK2-like catalytic subunit associated with plastid gene expression control. The protein is structurally CK2 alpha-like, comprising a conserved protein-kinase catalytic core (typical bilobal kinase architecture with an ATP-binding site and catalytic residues) and multiple conserved cysteine residues implicated in redox-sensitive regulation, consistent with its proposed role in integrating chloroplast redox state with phosphorylation-dependent signaling. Functionally, cpCK2 (also described in the literature as a plastid transcription kinase activity) phosphorylates plastid transcription machinery components and plastid sigma factors (e.g., SIG1/SIG6), thereby modulating promoter recognition and transcription by the plastid-encoded RNA polymerase system; phosphorylation can differentially influence sigma factor DNA-binding and promoter selectivity in vitro and is implicated in tuning chloroplast transcriptional outputs in vivo. More recently, the chloroplast-localized CK2 alpha subunit CPCK2 has also been shown to act in chloroplast-mediated defense regulation by phosphorylating the salicylic-acid-binding protein SABP3/carbonic anhydrase, promoting its S-nitrosylation and thereby dampening salicylic acid accumulation and innate immune responses, linking cpCK2-dependent phosphorylation to chloroplast redox/post-translational control pathways that impact nuclear-coordinated stress and immunity programs.
