EEF1D encodes a ubiquitously expressed subunit of the eukaryotic translation elongation factor 1 complex that localizes predominantly to the cytoplasm, with an alternatively spliced long isoform that can also function in the nucleus during cellular stress. The protein is produced as multiple isoforms: shorter forms that act within the elongation factor 1B (eEF1B) complex and a distinct long form generated by alternative splicing. Structurally, the shorter isoforms comprise the core elongation-factor subunit that associates with other eEF1 components, whereas the long isoform contains additional N-terminal sequence that confers stress-responsive, transcriptional regulatory activity. Functionally, the canonical role of EEF1D is as a guanine nucleotide exchange factor within the eEF1 complex, promoting recycling of eEF1A by facilitating GDP-to-GTP exchange to support delivery of aminoacyl-tRNAs to the ribosome during translation elongation; beyond this housekeeping function, the long isoform has been implicated in activating heat-shock element-containing genes as part of the heat shock response, and EEF1D is a demonstrated physiological substrate of protein kinase CK2, consistent with regulation by phosphorylation.
