Argonaute 3-PAZ encodes a cytoplasmic Argonaute-family protein that is enriched in germline perinuclear nuage and functions in small-RNA-guided silencing pathways. The protein exhibits the canonical Argonaute bilobal architecture, with an N-terminal lobe containing the PAZ domain (an OB-fold-like RNA-binding module that anchors the 3' end of bound small RNAs) and a C-terminal lobe comprising MID and PIWI domains (with MID contributing to 5'-end recognition and PIWI adopting an RNase H-like fold that can support "slicer" endonuclease activity in appropriate Argonautes). In Drosophila, Argonaute 3 acts as a core effector of the piRNA pathway, partnering with other PIWI-clade proteins in the ping-pong amplification cycle to process and amplify piRNAs and thereby repress transposable elements and help safeguard genome integrity in germ cells.
