PA1-1754 detects human beta-secretase 2 (BACE2). This product is a remanufactured version of PA1-754 produced in new rabbits.
PA1-1754 has been successfully used in Western blot procedures. By Western blot this antibody detects an ~58 kDa protein representing BACE2 in endogenous and overexpressing HEK-293 cells. PA1-1754 also detects two unidentified bands at ~61 kDa and ~80 kDa.
The PA1-1754 immunogen is a synthetic peptide corresponding to residues C P(503) E V V N D E S S L V R H R W K(518) of human BACE2. This sequence is completely conserved in mouse. The PA1-1754 immunizing peptide (Cat. # PEP-167) is available for use in neutralization and control experiments.
BACE2 or Beta Secretase 2 is responsible for the proteolytic processing of the amyloid precursor protein (APP). BACE2 cleaves APP between residues 690 and 691. This cleavage leads to the generation and extracellular release of beta-cleaved soluble APP. BACE2 is also know to cleave APP between residues 671 and 672. Accumulation of APP is a signature of Alzheimer disease and frequent complication of adult Down syndrome patients.
