IL-3Ra (interleukin-3 receptor alpha) is a member of the type I cytokine receptor family. It forms a heterodimer made of a unique alpha chain and a shared beta subunit (beta c/CD131). IL-3Ra transmits signals from interleukin-3 (IL-3). Binding of IL-3 promotes pairing of the alpha and beta chains, which is necessary for receptor activation, though not for high-affinity ligand binding. IL-3Ra is overexpressed on acute myeloid leukemia (AML) blasts compared with normal blood cells, making it a potential therapeutic target.
CD16, also called Fc gamma receptor III (FcgRIII), has two isoforms: FcgRIIIA (CD16A) and FcgRIIIB (CD16B), encoded by FCGR3A and FCGR3B on chromosome 1. FcgRIIIA is expressed on monocytes, macrophages, and natural killer (NK) cells, but not on T or B cells. It binds the Fc portion of IgG antibodies and helps clear immune complexes. When engaged, it triggers signaling pathways that promote antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, cytokine release, and antigen presentation. FcgRIIIB is a GPI-anchored receptor found mainly on neutrophils and supports similar immune responses, including cytotoxicity and enhanced viral interactions. Mutations in FCGR3A are linked to Immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, alloimmune neonatal neutropenia, and herpes zoster. Multiple transcript variants exist.
CD335 (NKp46/NCR1) is a natural cytotoxicity receptor expressed on NK cells. It is a type I transmembrane glycoprotein with two immunoglobulin-like domains. CD335 activates and expands resting NK cells, particularly through interactions with dendritic cells, and enhances their ability to kill tumor and virus-infected cells, including influenza-infected cells. Related to LIR and KIR receptors, CD335 plays a key role in tumor cell lysis and has also been implicated in autoimmune processes such as type 1 diabetes.
