Immunofluorescence analysis of Cyclin-dependent kinase 9 was performed using 70% confluent log phase A-431 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.

CDK9 is a serine/threonine kinase which when complexed with CyclinT1 forms an active holoenzyme complex. This complex plays a key role in regulating the eukaryotic cell cycle. CDK9 is coexpressed and copurified with CyclinT1. CDK9 regulates cytokine inducible transcription networks by facilitating promoter recognition of target transcription factors. CDK9 promotes RNA synthesis in genetic programs for cell growth, differentiation, and viral pathogenesis. The CDK9/cyclin-K complex also has kinase activity towards CTD of RNAPII and can substitute for CDK9/cyclin-T P-TEFb in vitro. Chronic activation of CDK9 causes cardiac myocyte enlargement leading to cardiac hypertrophy and confers a predisposition to heart failure.
