Carboxypeptidases A1, B1, and A2 (CPA1, CPB1, CPA2) are zinc-dependent exopeptidases produced as inactive zymogens in pancreatic acinar cells and activated in the intestinal lumen to complete protein digestion by trimming C-terminal amino acids from peptides. CPA1 and CPA2 preferentially remove hydrophobic or aromatic residues, while CPB1 removes basic residues like lysine and arginine, providing complementary substrate specificity to endopeptidases such as trypsin and chymotrypsin. These enzymes are encoded by distinct genes with high expression in the pancreas, secreted into the duodenum via the pancreatic duct, and stabilized by cofactors and pH conditions optimal for digestive function. Clinical relevance includes CPA1 variants linked to early-onset pancreatitis through misfolding stress, CPB1 as a biomarker in pancreatic injury, and altered carboxypeptidase activity in malabsorption and inflammatory states.
