Specific Species Reactivity: Chlamydomonas reinhardtii
CrPDAT1 encodes a phospholipid:diacylglycerol acyltransferase of the LCAT-like family in the unicellular green alga Chlamydomonas reinhardtii; the protein is synthesized as a soluble, N-terminally targeted enzyme proposed to act at the chloroplast based on the presence of a transit peptide, and recombinant CrPDAT retains activity when its N-terminus is truncated, consistent with a modular organization containing a catalytic core typical of LCAT-like acyltransferases. Functionally, CrPDAT1 catalyzes acyl-CoA-independent triacylglycerol (TAG) formation by transferring an acyl group from membrane phospholipids and galactolipids to diacylglycerol, and it can also mediate DAG:DAG transacylation, thereby diverting fatty acid flux from membrane lipids into storage TAG. In addition to acyltransferase activity, CrPDAT1 exhibits broad lipase/acyl-hydrolase activities toward TAG and multiple membrane lipids, supporting a role in coordinated membrane lipid turnover and remodeling. Genetic perturbation reduces TAG accumulation during nitrogen starvation and alters membrane lipid composition with impaired growth, indicating CrPDAT1 is important both for robust proliferation in favorable conditions and for stress survival by coupling membrane lipid degradation to TAG production.
