MX823 specifically reacts with drebrin, a widespread actin-associating protein of 70 kD (SDS-PAGE mobility shows MW 120,000). It is enriched at junctional plaques, defining a specific microfilament anchorage system in polar epithelial cells. Positive staining was found in many tissues, including diverse epithelia and carcinoma (e.g. in epidermis basal cells are positive, whereas overlying cells are negative; also positive: regenerating epithelium during wound healing; basal cell carcinoma; skin melanoma), specific types of endothelia and smooth muscle; especially prominent in the mesangial cells of renal glomeruli and in the Sertoli cells of testis. Also positive: epithelia of hair follicles and eccrine sweat glands. Consistently negative were, however, hepatocytes and cross-striated muscle.Reactivity on Cultured cell lines: Primary human keratinocytes, U333, MCF-7, SV80; B16 (mouse melanoma cells).
The protein encoded by this gene is a cytoplasmic actin-binding protein thought to play a role in the process of neuronal growth. It is a member of the drebrin family of proteins that are developmentally regulated in the brain. A decrease in the amount of this protein in the brain has been implicated as a possible contributing factor in the pathogenesis of memory disturbance in Alzheimer's disease. At least two alternative splice variants encoding different protein isoforms have been described for this gene.
