FtsH3 and FtsH10 encode closely related, nuclear-encoded AAA+ zinc metalloproteases of the FtsH/M41 family that localize to plant mitochondria, where they are embedded in the inner mitochondrial membrane and assemble into high-molecular-weight m-AAA protease complexes. Each protein is synthesized with an N-terminal mitochondrial targeting region followed by membrane-anchoring segments and a large catalytic moiety containing a conserved AAA ATPase (unfoldase/chaperone) domain coupled to a HEXXH zinc-dependent protease domain that together drive ATP-dependent extraction, unfolding, and proteolysis of membrane-embedded substrates. Functionally, the FtsH3/FtsH10 m-AAA system contributes to mitochondrial proteostasis by supporting quality control and remodeling of oxidative phosphorylation components; genetic and biochemical evidence indicates FtsH3 activity influences respiratory Complex I abundance by promoting complex disassembly/turnover, and FtsH3 and FtsH10 have been implicated in mitochondrial translation-related protein processing, consistent with a broader role in maintaining inner-membrane protein homeostasis under basal and stress conditions.
