FtsH6 encodes a nuclear-encoded, chloroplast-localized ATP-dependent zinc metalloprotease that associates with the thylakoid membrane, where it contributes to plastid protein quality control and regulated turnover of specific substrates. The FtsH6 protein is a membrane-anchored member of the FtsH family, characterized by an N-terminal transmembrane segment that tethers the enzyme to the membrane, followed by a soluble region containing an AAA+ ATPase module (supporting ATP-driven substrate unfolding/translocation) and a C-terminal M41 metalloprotease domain harboring the conserved zinc-binding HExxH motif required for proteolysis. Functionally, genetic evidence in Arabidopsis shows FtsH6 is required for efficient degradation of light-harvesting complex II components (including Lhcb proteins) during high-light acclimation and during dark-induced senescence, consistent with a role in remodeling photosynthetic antenna composition under changing conditions. In addition, FtsH6 has been implicated in regulating heat-stress thermomemory by promoting the decline of the plastid small heat shock protein HSP21 during the post-stress memory phase, thereby tuning the duration of acquired thermotolerance.
