Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is brain tissue.
G-rich sequence factor 1 (GRSF1) is a regulator of post-transcriptional mitochondrial gene expression. It required for assembly of the mitochondrial ribosome and for recruitment of mRNS and IncRNA. GRSF1 binds RNAs containing the 14 base G-rich element. It preferentially binds RNAs transcribed from three contiguous genes on the light strand of mtDNA, the ND6 mRNA, and the long non-coding RNAs for MT-CYB and MT-NDS - each of which contains multiple consensus binding sequences.
