H4K12su Antibody (PA5-112553) in ICC/IF
Immunocytochemichal analysis of Succinyl-Histone H4 (Lys12) in Hela cells using a Polyclonal antibody (Product # PA5-112553) at a dilution of 1:5. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated Goat Anti-Rabbit IgG(H+L). Cells were also counter-stained with DAPI.

H4K12su is the SUMOylation of lysine 12 on histone H4, a core nucleosomal protein (UniProt histone H4 family) involved in chromatin structure. This modification is associated with transcriptional repression by inhibiting activating marks like acetylation and H3K4 methylation. Despite promoting a more open chromatin structure, it functions as a regulatory epigenetic mark that fine-tunes gene expression.
