Immunocytochemistry/Immunofluorescence analysis of HERC5 in U2OS cells using HERC5 Polyclonal Antibody (Product # PA5-114382). Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum and incubated with the primary antibody at 5 µg/mL. Positive Control - WB: human 293T whole cell, human A549 whole cell, human PC-3 whole cell. ICC/IF: U2OS cell.|Store at -20°C for one year from date of receipt. HERC5 (E3 ISG15 - protein ligase HERC5) is a major E3 ligase for ISG15 conjugation. It acts a positive regulator of innate antiviral response in cells induced by interferon. HERC5 functions as a part of the ISGylation machinery that recognizes target protein in a broad and relatively non-specific manner. It catalyzes ISGylation of IRF3 - which results in sustained activation, it attenuates IRF3-PIN1 interaction - which antagonizes IFR3 ubiquitination and degradation, and boosts the antiviral repsonse. HERC5 is physically assocated with polyribosomes and broadly modifies newly synthesized proteins in a cotranslational manner.
