Maduramicin is a potent polyether antibiotic primarily used as an anticoccidial agent in veterinary medicine, particularly in poultry. It functions by disrupting ion transport across cell membranes, which is crucial for its efficacy against coccidia. Recent studies have explored its potential repurposing as an anticancer agent, particularly against triple-negative breast cancer (TNBC), due to its ability to inhibit calcium channel activity and alter cellular migration. The biosynthetic gene cluster responsible for maduramicin production has been identified, providing insights into its genetic and molecular structure, although genetic engineering of associated strains remains a challenge. Maduramicin's structure allows it to effectively bind and transport ions, which is central to its mechanism of action. Despite its benefits, maduramicin poses significant toxicity risks, leading to fatal poisonings in non-target species, including humans.
