Neuregulin-1 (NRG1) is a gene encoding multiple isoforms involved in various physiological functions, particularly within the nervous system. The SMDF (sensory and motor neuron-derived factor) is one such isoform of NRG1. Located on chromosome 8p12, NRG1 comprises several exons, where alternative splicing gives rise to the diverse isoforms. The SMDF isoform of NRG1 plays a critical role in neural development and the maintenance of neuronal functions, including neurotransmission and synaptic plasticity. This isoform signals through interactions with ErbB family receptors, predominantly ErbB3 and ErbB4, facilitating cellular communication necessary for proper neural function. NRG1, including its SMDF variant, has been implicated in neuropsychiatric conditions such as schizophrenia and bipolar disorder, underscoring its significance in both development and disease.
