Nosiheptide is a potent thiopeptide antibiotic characterized by a macrocyclic core containing a 6-membered heterocycle central to multiple azoles and dehydroamino acids. It is produced through ribosomal synthesis followed by post-translational modifications (PTMs) of precursor peptides, which contribute to its structural complexity. The biosynthesis of nosiheptide involves a class C radical S-adenosylmethionine methylase, NosN, which catalyzes carbon transfer to C4 of 3-methylindolic acid, completing the side-ring system. Nosiheptide exhibits excellent bactericidal activity against Gram-positive bacteria, although its physical and chemical properties limit its clinical application. Research continues to explore its biosynthetic pathway and potential analogues for antibacterial use.
