PIF3 is a nuclear-encoded transcription factor that accumulates in the nucleus and functions as a direct signaling partner of photoactivated phytochromes to couple light perception to transcriptional reprogramming during seedling development. The encoded protein is a basic helix-loop-helix regulator with a C-terminal bHLH DNA-binding/dimerization domain and an N-terminal PAS (Per-Arnt-Sim-like) region that supports protein-protein interactions, consistent with its role as a phytochrome-interacting factor. Functionally, PIF3 binds G-box-type cis elements in promoters of light-regulated genes and acts as a key modulator of photomorphogenesis and chloroplast biogenesis, contributing to rapid light-induced expression of nuclear genes needed for establishment of the photosynthetic apparatus at the dark-to-light transition while also repressing chlorophyll biosynthesis/photosynthesis programs in darkness through recruitment of chromatin repressors such as the histone deacetylase HDA15.
