PTCH2 Antibody — Figure 3. Abeta 1-42 not Abeta 1-40 increases the expressions of Shh signaling in hippocampal GPCs derived from WT mice. The suspended cells were incubated with the presence of 0, 0.1, 1, 10 or 100 u m of Abeta 1-42 or Abeta 1-40 for 3 days. ( A ) Shh expression with different molecular weights was shown by western blot from the cultured medium and the cells treated with Abeta 1-42 . ( B ) Shh levels were significantly increased in the conditioned medium (normalized to corresponding cell lysate) and cultured cells in a dose-dependent manner with Abeta 1-42 treatment (ANOVA test, * P < 0.05, **P < 0.01). ( C ) The expression of Shh signaling components was shown in the cells with Abeta 1-42 treatment. ( D ) The levels of Ptc1, Ptc2, Smo, Gli1 and Gli2 were significant elevated (ANOVA test, * P < 0.05, * * P < 0.01) but not significant changes of Gli3 levels with the exposure of Abeta 1-42 (ANOVA test, P > 0.05). ( E ) Shh expression was shown from the conditioned medium and GPCs treated with Abeta 1-40 . ( F ) Shh levels were not significantly increased in the conditioned medium (normalized to corresponding cell lysate) and the cultured GPCs with the Abeta 1-40 treatment (ANOVA test, P > 0.05). ( G ) The expression of Shh signaling component molecules was shown in the GPC lysate with the Abeta 1-40 treatment. ( H ) The levels of the components were not significantly changed with the Abeta 1-40 treatment. ( I ) Western blot showed Ptc1 expression with the exogenous Shh applicat {TM}
