RAF2 is a plant chloroplast protein that functions as a Rubisco assembly factor, promoting proper folding and integration of Rubisco large and small subunits to form an active carboxylase for CO2 fixation. It cooperates with chaperones and assembly partners such as RAF1, RbcX, and chaperonins to stabilize intermediates and ensure efficient biogenesis of Rubisco during chloroplast development and under changing environmental conditions. By supporting Rubisco assembly, RAF2 impacts photosynthetic capacity, redox balance, and growth, and its expression aligns with high photosynthetic demand and light regulated chloroplast maturation. Perturbation of RAF2 reduces Rubisco accumulation and activity, lowering carbon assimilation and biomass, while coordinated regulation of RAF2 helps plants maintain photosynthetic efficiency and recover from stress that challenges protein assembly in the thylakoid and stroma.
