Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

RGA1 encodes a cytoplasmic Rho-family GTPase-activating protein that concentrates transiently at cortical division-site landmarks (e.g., the bud neck/bud scars) and can redistribute to the cytoplasm under replication stress, consistent with a role in spatial control of polarity signaling. The protein is modular, featuring tandem N-terminal LIM zinc-binding domains (protein-interaction/scaffolding modules) followed by a C-terminal RhoGAP catalytic domain that stimulates GTP hydrolysis on its target GTPase. Functionally, RGA1 negatively regulates the pheromone-response MAPK pathway and polarized morphogenesis by down-modulating the activity of the polarity GTPase Cdc42, thereby contributing to proper bud-site selection and septin organization and preventing inappropriate Cdc42 repolarization after cytokinesis.
