Western blot analysis of SENP1 using SENP1 antibody (Product # MA5-49217). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 µg of sample under reducing conditions. SENP1 is a protease that catalyzes two essential functions in the SUMO pathway: processing of full-length SUMO1, SUMO2 and SUMO3 to their mature forms and deconjugation of SUMO1, SUMO2 and SUMO3 from targeted proteins. SENP deconjugates SUMO1 from HIPK2 and from HDAC1, which decreases the transcriptional repression activity of the latter.
