TGA1 encodes a nuclear, sequence-specific DNA-binding transcription factor of the basic region/leucine zipper (bZIP) family that functions as a dimer and is enriched for an N-terminal regulatory region coupled to a C-terminal basic DNA-binding region followed by a leucine-zipper dimerization interface; consistent with bZIP proteins, the basic region confers recognition of ACGT-containing promoter elements, and TGA1 displays strong preference for TGACG-centered motifs (including ocs/as-1-type elements) that occur in defense- and hormone-responsive promoters. In Arabidopsis, TGA1 participates in transcriptional reprogramming during salicylic acid-associated immunity and systemic acquired resistance by binding TGACG/as-1-like cis elements and cooperating with the master cofactor NPR1, with NPR1 recruitment and/or activity linked to the redox state of conserved cysteine residues in TGA1 that modulate NPR1 interaction and downstream promoter activation.
