TIP2 is a plant tonoplast intrinsic protein belonging to the aquaporin family that facilitates movement of water and small neutral solutes such as ammonia across the vacuolar membrane. By regulating vacuolar water permeability, TIP2 helps control cell turgor, osmotic balance, and rapid volume changes during growth, stomatal function, and stress acclimation. Some TIP2 isoforms conduct ammonia or hydrogen peroxide, linking vacuolar transport to nitrogen metabolism and redox signaling. Expression of TIP2 is tissue specific, often strong in roots and developing tissues, and responds to drought, salinity, and hormone cues, with gating influenced by pH, phosphorylation, and membrane tension. Perturbation of TIP2 alters vacuolar dynamics, osmotic adjustment, and stress tolerance, highlighting its role in water relations and metabolic homeostasis.
