The gene produces a multi-exon transcript whose protein product is characterized by an N-terminal receiver-like (pseudo-receiver) domain typical of response regulators but lacking canonical phospho-acceptor activity, and a C-terminal CCT (CONSTANS, CO-like, TOC1) DNA-binding region required for direct promoter occupancy and repression of central clock genes such as CCA1 and LHY; deletion or disruption of the CCT region abolishes DNA binding and impairs repression. TOC1-P activity is further tuned by post-translational regulation, including N-terminal phosphorylation that modulates interactions with specific transcriptional regulators at rhythmic promoters, and protein-protein interactions that influence nuclear accumulation and stability, thereby coupling clock timing to photoperiodic outputs such as growth and flowering and to environmental responses including temperature-gated elongation programs.
