X-box binding protein 1, spliced form (XBP1S), is a critical transcription factor involved in the unfolded protein response (UPR). Located on chromosome 22q12, XBP1S is derived from the unspliced XBP1 mRNA through a unique splicing event mediated by the endoribonuclease IRE1 alpha under endoplasmic reticulum (ER) stress conditions. Structurally, XBP1S belongs to the cAMP-response element-binding (CREB)/activating transcription factor (ATF) family and contains a basic-region leucine zipper (bZIP) domain that enables its function as a transcription factor. Functionally, XBP1S migrates to the nucleus where it regulates a broad range of genes involved in ER-associated degradation (ERAD), lipid biosynthesis, and molecular chaperones, aimed at restoring homeostasis and decreasing ER stress. Additionally, the IRE1 alpha-XBP1S signaling pathway has been implicated in cancer cell proliferation and various diseases characterized by misfolded protein accumulation.
