Aminoglycoside adenyltransferase (AadA; aminoglycoside O-nucleotidyltransferase) is a bacterial cytosolic enzyme typically encoded by horizontally mobile antibiotic-resistance determinants (commonly plasmid- and integron-associated) and produced as a soluble monomeric protein. Structurally, AadA adopts a two-domain architecture comprising an N-terminal nucleotidyltransferase (NTase) catalytic domain and a C-terminal alpha-helical bundle domain; the active site lies in an interdomain cleft and undergoes ligand-coupled conformational rearrangements, with ATP binding occurring prior to aminoglycoside binding to properly position the domains for catalysis. Functionally, AadA mediates aminoglycoside resistance by catalyzing a Mg2+-dependent transfer of AMP from ATP onto hydroxyl groups of target antibiotics, including O-adenylation at the 3''-hydroxyl of streptomycin and the 9-hydroxyl of spectinomycin, thereby reducing drug affinity for the ribosome and diminishing inhibition of protein synthesis; mutational and structural studies support a key role for a conserved glutamate (e.g., Glu87 in Salmonella enterica AadA) as the catalytic base and identify basic residues important for ATP coordination during the adenylation reaction.
