HHF35 reacts with both alpha-muscle and gamma-smooth muscle actin, and therefore reacts with skeletal muscle, cardiac muscle, vascular and visceral smooth muscle cells, pericytes and myoepithelial cells. It is also reactive in myofibroblasts. It does not react with epithelial, endothelial, neural or normal connective tissue cells when applied under the proper conditions to these tissue sections.
The epitope recognized by α-SM1 is highly conserved. The antibody therefore cross-reacts with many species including protochordates, lower craniates and mammals.
The antibody may be stored at 4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at 4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
Actins are highly conserved proteins that are involved in various types of cell motility, and maintenance of the cytoskeleton. In vertebrates, three main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins co-exist in most cell types as components of the cytoskeleton, and as mediators of internal cell motility. Actin gamma 1 is a cytoplasmic actin found in nonmuscle cells.
