Reconstitute the lyophilized antiserum by adding 1 mL sterile distilled water.
Inter-species cross-reactivity is a normal feature of antibodies to Serum proteins, since homologous proteins of different species frequently share antigenic determinants. Crossreactivity of this antiSerum has not been tested in detail. In double radial immunodiffusion a reaction has been observed with Bovine, Cat, Dog and Monkey.
In immunoelectrophoresis use 2 µL serum or equivalent against 120 µL antiserum. In double radial immunodiffusion use a rosette arrangement with 10 µL antiserum in 3 mm diameter centre well and 2 µL serum samples (neat and serially diluted) in 2 mm diameter peripheral wells. In electroimmunodiffusion the amount of antiserum required in the agarose gel is usually between 1 and 2%, depending on the test arrangement. The amount of platelet factor 4 precipitated by 1 mL antiserum is approximately 0.1 mg.
Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at 4°C, not refrozen, and preferably used the same day.
Platelet factor-4 is a 70-amino acid protein that is released from the alpha-granules of activated platelets and binds with high affinity to heparin. Its major physiologic role appears to be neutralization of heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation. As a strong chemoattractant for neutrophils and fibroblasts, PF4 probably has a role in inflammation and wound repair (Eisman et al., 1990).
