The Bovine/Sheep Cortisol ELISA quantitates Cortisol in serum and plasma
原理方法:
The Competitive Cortisol ELISA research-use-only kit is designed to quantitatively measure Cortisol in Bovine and Sheep. A Cortisol standard is provided to generate a standard curve for the assay and all samples are read off a user-generated standard curve. Standards or diluted samples are pipetted into a coated microtiter plate and Biotinylated Detection Ab specific to Bovine/Sheep Cortisol is added. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured on a microplate reader. The concentration of Bovine/Sheep Cortisol in the samples is then determined by comparing the OD of the samples to the standard curve

Cortisol is the most potent glucocorticoid produced by the human adrenal. It is synthesized from cholesterol and its production is stimulated by pituitary adrenocorticotropic hormone (ACTH) which is regulated by corticotropin releasing factor (CRF). ACTH and CRF secretions are inhibited by high cortisol levels in a negative feedback loop. In plasma a majority of cortisol is bound with high affinity to corticosteroid binding globulin (CBG or transcotin). Cortisol acts through specific intracellular receptors and affects numerous physiologic systems including immune function, glucose counter regulation, vascular tone, and bone metabolism.
