The Human HTRA1 (HtrA Serine Peptidase 1) ELISA quantitates HTRA1 in serum and plasma.
原理方法
The Human HtrA Serine Peptidase 1 (HTRA1) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of HtrA Serine Peptidase 1 (HTRA1) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to HtrA Serine Peptidase 1 (HTRA1). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of HtrA Serine Peptidase 1 (HTRA1) present in the original specimen.

HtrA1, a 51KDa protein, is a member of highly conserved mammalian HtrA serine protease family. It is a human homolog of bacterial HtrA and consists of a secretory signal sequence, an IGFBP-binding domain and a Kazal-type serine-protease inhibitor domain at the N-terminus and a highly conserved trypsin-like serine/protease domain and a PDZ domain at the C-terminus. It plays a major role in the regulation of cell growth. HtrA1 inhibits signaling mediated by TGF-beta family members and regulates many physiological processes. HtrA1 is up-regulated in placenta during pregnancy and plays an important role in implantation and placental physiology. It regulates the availability of IGFs by cleaving IGF-binding proteins. It acts as a tumor suppressor and is down-regulated in ovarian cancers and melanomas. Its role has been implicated in osteoarthritis suggesting a therapeutic intervention. Northern blot analysis detected high expression in placenta.
