The Mouse MMP-3 (Matrix Metalloproteinase 3) ELISA quantitates MMP-3 in serum and plasma.
Principle of the method
The Mouse Matrix Metalloproteinase 3 (MMP-3) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the concentration of Matrix Metalloproteinase 3 (MMP-3) in biological samples. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody.
The sandwich complex is formed by the addition of a second (HRP-linked) detection antibody specific to Matrix Metalloproteinase 3 (MMP-3). Excess reagents are washed from the plate.
A substrate solution is added that reacts with the enzyme-antibody-target complex to produce a measurable signal. The enzyme-substrate reaction is terminated by the addition of stop solution. The intensity of the signal, measured spectrophotometrically at 450 ± 2 nm, is directly proportional to the concentration of Matrix Metalloproteinase 3 (MMP-3) present in the original specimen.

MMP3 (stromelysin-1) belongs to the peptidase M10A subfamily. It is secreted in its pro-protein form, and is activated by extracellular proteinase cleavage. MMP3 is capable of binding to zinc and calcium ions, and has metallo-endopeptidase activity. The active enzyme degrades collagen, gelatin, fibronectin, and laminin. It is involved in normal cellular processes such as wound repair and tissue remodeling. Mutations in the gene can result in coronary heart disease 6.
