This workshop is designed to help you accelerate your sample preparation for cryo-electron microscopy (cryo-EM). Lectures will provide insights on overcoming challenges in protein biochemistry, the sample vitrification process and procedure, and hands-on imaging in both room- and cryo-temperature electron microscopes to test different sample preparation parameters.

Due to limited space, we are allowed only 10 participants. You will be notified by email on the status of your registration.

Monday, May 27th

10:00 – 10:45 a.m.	Welcome and Introduction to CryoEM at Molecular Horizons (James Bouwer)
11:00 – 11:45 a.m.	The Sample Preparation Workflow and Techniques (Shiny Shi)
12:00 – 1:00 p.m.	Lunch
1:30 – 4:00 p.m.	Hands-on dry run on sample preparation
5:00 – 7:00 p.m.	Dinner (Location TBD)

Tuesday, May 28th

12:00 – 1:00 p.m.	Lunch
1:15 – 4:00 p.m.	Continue hands-on sample preparation thru cartridge loading into cassette
5:00 – 7:00 p.m.	Dinner (Location TBD)

10:00 a.m. – 12:00 p.m.	Hands-on sample preparation on negative stain
12:00 – 1:00 p.m.	Lunch
1:15 – 4:00 p.m.	Continue hands-on sample preparation on negative stain and imaging

Basic sample requirements for room- and cryo-temperature imaging in electron microscopy.
Factors that help achieve optimal homogeneity, concentration, orientation and ice thickness.
Standard operating procedures and review of additional factors/options on grid selection and optimization.
Hands-on sample preparation with facility equipment.

Tuesday, May 28: Hands-on imaging workshop

Negative stain imaging at 120 kV to test sample prep protocols.
Discussion of outcomes.

Wednesday May 29: Hands-on imaging workshop

Using 200 kV Thermo Scientific™ Talos Arctica Cryo-TEM to test sample prep and vitrification protocols.
Setup of basic imaging to test sample prep protocols.
Group presentation of room-temperature micrograph images and discussion.
Thursday, May 30 (AM): More hands-on with 200 kV Cryo-TEM
Group presentations of cryo micrograph images and discussion.

Thursday, May 30 (PM): Molecular Horizons

Ongoing cryo-EM service lab facility options and roadmap.
Thermo Fisher Scientific presentation on Integrated Structural and Cellular Biology Workflows.
