The ADPGK gene encodes ADP-dependent glucokinase, an enzyme distinct from the ATP-dependent hexokinases as it employs adenosine diphosphate (ADP) rather than ATP to phosphorylate glucose to glucose-6-phosphate. This gene, initially identified in archaeal species, has been co-opted into mammalian systems where it plays roles primarily in glycolytic processes. Structurally, the mammalian ADPGK differs from its archaeal counterpart, exemplified by the rearrangement of its secondary structures, such as the alpha2 and alpha3 helices. These helices are replaced by an extended beta-strand region, indicating evolutionary adaptations for function in higher organisms like mice. Although its precise physiological role remains partially defined, ADPGK is notably expressed in both hematopoietic lineages and tumor tissues, suggesting potential significance in cancer biology and energy metabolism.
