CD43 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of CD43 was performed using 70% confluent log phase U-937 and HT-29 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with CD43 Monoclonal Antibody (SP55) (Product # MA5-16339) at 1:100 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, Alexa Fluor 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image of U-937 showing plasma membrane and cytoplasm localization. Panel e represents HT-29 cells having no expression of CD43. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

CD43 Antibody in Immunohistochemistry (IHC) — Immunohistochemical analysis of CD43 using anti-CD43 Monoclonal Antibody (Product # MA5-16339) in Tonsil Tissue. The recommened dilution for this antibody in immunohistochemistry applications is 1:200.

CD43 Antibody in Western Blot (WB) — Western blot was performed using Anti-CD43 Monoclonal Antibody (Product # MA5-16339) and a 110 kDa band corresponding to CD43 was observed across the cell lines tested. Membrane enriched extracts (30 µg lysate) of MOLT-4 (Lane 1), HEL 92.1.7 (Lane 2), U-937 (Lane 3) and HT-29 (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:250 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

CD43 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines tested owing to their inherent genetic constitution. Relative expression of CD43 was observed in U-937 cells with membranous localization, in comparison to HT-29, a negative cell line for CD43, using Anti-CD43 Monoclonal Antibody (Product # MA5-16339) in Immunofluorescence (DOI: 10.1016/j.yexmp.2011.10.009). {RE}

Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is tonsil tissue.

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with high negative charge, expressed on the surface of most hematopoietic cells. CD43 contributes to a repulsive barrier that interferes with cellular adhesion, however, in certain cases also promotes leukocyte aggregation. By interaction with actin-binding proteins ezrin and moesin CD43 plays a regulatory role in remodeling T-cell morphology and regulates cell-cell interactions during lymphocyte traffic. CD43 signaling both enhances LFA-1 adhesiveness and counteracts LFA-1 induction via other receptors. Expression of CD43 causes induction of functionally active tumor suppressor p53 protein, but in case of p53 and ARF deficiency CD43 promotes tumor proliferation and viability. CD43 appears to be an important modulator of leukocyte functions. Diseases associated with CD43 dysfunction include Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
