Immunofluorescence analysis of CNPase was performed using 70% confluent log phase Hep G2 and MCF7 cells. CNPase Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of CNPase was performed using 70% confluent log phase Hep G2 and MCF7 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with CNPase Polyclonal Antibody (Product # PA5-86561) at 1:100 in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor™ Plus 488 (Product # A32790, 1:2,000), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b: Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing Plasma Membrane and with minimal cytoplasm localization in Hep G2 cells. Panel e represents MCF7 with no signal. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

CNPase Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of CNPase in paraffin-embedded human colorectal carcinoma tissue. Sample was incubated with polyclonal CNPase antibody (Product # PA5-86561) using a 1:50 dilution.

CNPase Antibody in Western Blot (WB) — Western blot was performed using CNPase Polyclonal Antibody (Product # PA5-86561) and a -45 kDa band corresponding to CNPase was observed across all cell lines and tissues. Membrane enriched extracts (30 µg lysate) of MOLT-4 (Lane 1), Hep G2 (Lane 2), SK-BR-3 (Lane 3), HeLa (Lane 4), MCF7 (Lane 5), Mouse Brain (Lane 6), Mouse Cerebellum (Lane 7), Rat Brain (Lane 8), Mouse Lung (Lane 9), Mouse Colon (Lane 10) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).

CNPase Antibody in Western Blot (WB) — Western blot analysis of CNPase in Lane 1: THP-1 whole cell lysate (40 µg), Lane 2: SGC7901 whole cell lysate (40 µg), Lane 3: CT26 whole cell lysate (40 µg), Lane 4: PC12 whole cell lysate (40 µg). Sample was incubated with CNPase polyclonal antibody (Product # PA5-86561) using a 1:500 dilution.

CNPase Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines and tissues owing to their inherent genetic constitution. Relative expression of CNPase was observed in MOLT-4, Hep G2, SK-BR-3, HeLa, Mouse Brain, Mouse Cerebellum and Rat Brain compared to MCF-7, Mouse Lung and Mouse Colon which are reported as negative models using CNPase Polyclonal Antibody (Product # PA5-86561) in Western Blot. {RE}

CNPase Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell models owing to their inherent genetic constitution.
