DRB4 is a protein-coding gene within the major histocompatibility complex class II region and encodes an HLA-DR beta chain that pairs with the invariant HLA-DRA alpha chain to form a cell-surface, membrane-anchored heterodimer on professional antigen-presenting cells. The encoded beta chain is synthesized as a type I membrane glycoprotein precursor (approximately 26-28 kDa) with an N-terminal signal peptide, two extracellular domains that create the peptide-binding platform and Ig-like domain, a single-pass transmembrane helix, and a short cytoplasmic tail; gene organization classically places the leader sequence in exon 1, the extracellular domains in exons 2-3, the transmembrane segment in exon 4, and the cytoplasmic tail in exon 5 (with additional untranslated or alternatively annotated exons reported on alternate loci). Functionally, the DRB4-containing HLA-DR complex binds peptides generated mainly in the endocytic/lysosomal pathway from extracellular or vesicular proteins and displays them at the cell surface for surveillance by CD4 T cells, thereby shaping adaptive immune responses; extensive allelic polymorphism in the beta chain is a key determinant of peptide-binding specificity and underlies its importance in histocompatibility matching and disease association studies.
