The EPEA tag also known as the C-tag is a short peptide sequence consisting of the four amino acids glutamic acid-proline-glutamic acid-alanine (E-P-E-A) used as an affinity tag for protein purification and detection. This tag is strategically placed at the C-terminus of recombinant proteins and is primarily utilized in affinity chromatography to facilitate efficient and convenient purification processes. The EPEA tag binds specifically to capture antibodies or resins enabling selective retrieval of the tagged protein from a complex mixture without impacting protein function. Unlike more commonly used tags like His-tag the EPEA tag is less likely to interfere with protein folding and activity providing an advantage in maintaining the biological activity of tagged proteins during purification. It is commonly employed in biotechnology and research laboratories for applications requiring highly pure protein samples such as structural studies functional assays and therapeutic protein production.
